I know exactly where you are talking about - I pass it all the time but NEVER seen the Pacific. Its probably a discreet little entrance - like the adult bookshops and brothels in that same block .
Nah you'll be right. Whilst it aint the nicest part of town its VERY busy so you cant drop a smoke bomb and just go missing off the face of the earth.
And yep not worth the one night thing. Two gives you a bit more time to look around. Take the ferry over to the zoo one day - thats nice

If you're staying two nights.. i'd recommend the Maestri Towers.... i had a great place that overlooked darling harbour on the powderchair/Snow Patrol APEC weekend heh... it was the Penthouse Suite though... so maybe i can't compare the other rooms with that haha
but i just saw it on Wotif.. for a reasonable Wed/thurs, but i shall check out their own website for you as sometimes that's cheaper anyway!

When I went to Melb by phone booking they wanted to charge me $230 a night online it was $135 and we had the option of late checkout or breaksfast.
We took the breaksfast it wouldve cost us $60 for brekkies so was a good bonus...:)
