I don't believe that he is "often" asked that. American's general view of Australia probably isn't accurate (it's likely a mashup of Crocodile Dundee, Crocodile Hunter, Mad Max and Men At Work videos) but I don't know of anybody who thinks Australians don't speak English.
Unless, as somebody mentioned, it's just trash talk.
I'm an Aussie living In the US and I have got asked what language I spoke when I was in Australia several times... It's demeaning and had no idea people can be that sheltered not to know simple things like that.

It's possible somebody said it sincerely, after all there are stupid people in the world, and most people can't hear their own accent. But I think maybe they were just having a little fun with your uncle.
