What about biggest non festival where there was only one stage? I don't know the numbers but if I remember right some of the South American shows were in the 50,000 range.
What about somewhere in South America? Those soccer (or football if you prefer) stadiums down there hold upwards of 75,000 people.

As for Lolla, most people there seem to agree that Lolla takes the crown with estimates of 100,000 or more, though officially Billboard reported the attendance for the weekend at 168,729 (56,243 each day if you divide by 3) from a 200,000 (66,666) capacity with no days sold out. Another Billboard an article from earlier this year (when Radiohead played?) stated "For the first time since its reinvention as a destination festival in 2005, a single day of Lollapalooza sold out at 75,000 tickets. Tickets are also gone for today but remain for Sunday." So seems must've been slightly less for Pearl Jam in 07.
I guess we may never no but it can't hurt to speculate. So here's some other notable big attendances.

I heard upwards of 80,000 but it could be more, there were people stretched the length of grant park.

Palacio de los Deportes in Mexico City has approximately 22,000 seats, of course this does not include the floor area, but still I think it would be hard pressed to beat out other venue's already listed.
As for Lolla, the field was completely packed, there may have been a lot of invited guests that were not official ticket holders, but that section of grant park was packed. Compared to radio head this year, which sold out at 75,000 tickets, I would wager there was more attendance at Pearl Jam last year.

I guess we'll never know as all the eye witness accounts pretty much double it. As for other festivals and multiple stages, I think at least for the last hour at Reading/leeds the only stages open are the main stage and the 2nd stage (tent) which holds about 5,000, again of course you'll never get 100% acurate stats on how many attended Pearl Jam or whoever.

someone posted a day or two after the concert saying they talked to some cops that night and they said they figured it was 115,000 to 125,000 in grant park

Nijmegen was the biggest PJ show that I've been part of; 40,000 sounds about right. I was about 6 rows back and looking behind me was scary.
Nijmegen was a big crowd but a friendly crowd, i was 2nd row and it was not rough at all and only very occasionally was it even pushy
