The Hot Rock is awesome, and my favorite...but I think you may be best starting with their most recent...the title escapes me at the moment. I think Sleater-Kinney is kind of an acquired taste as far as their sound...most people I've showed them to just don't get it and find Corin highly annoying.
Edit: Their most recent is The Woods. Hopefully you end up liking them, they have a good amount of material to enjoy.

Depends on what you like to listen to already. Personally, Dig Me Out is killer but it's pretty aggressive and they weren't into harmonizing on that one. I'd recommend All Hands on the Bad One like someone else did above. Personally I think it's their best one. In my opinion it's where they finally had their "a-ha" moment about understanding what role harmonies could do for them. And, lets face it, there aren't too many bands these days that are capable of harmonizing as well as they did on this one.
The Hot Rock had a few good cuts on it, but I don't think it was their strongest all around effort.

they're all good in their own way, and each release has a distinct sound and feel to it.
The Woods, their last, has a big rock sound, and some of their best songs (Jumpers, Rollercoaster, Entertain, Modern Girl, Lets Call it Love). It's raucous and loud.
Dig Me Out was the album that broke them, and it amazing all the way through.
One Beat is probably my favorite. It takes a lot of the interesting vocal harmonies they were doing on The Hot Rock but beefs them up. It's also the album after Corin had her baby, and the anti Bush album. But it's not preachy or pedantic at all. (One Beat, Light Rail Coyote, Sympathy, Far away, Combat Rock are my favorites).
I have been a fan of theirs since the beginning, so it's nice to see people just getting into them now!

"Dig Me Out" is my favorite album of their's......then "One Beat".

The Hot Rock is my personal fave but I would agree with All Hands On the Bad one as starting point...I think it's the easiest to get into. Enjoy whichever you choose!

Another SK fan here, it's hard to say which is my favorite, but i'd suggest starting with Dig Me Out and working your way towards The Woods. The records before that are okay, but in my opinion, SK didn't become totally badass until Janet joined. Oh and btw, Carrie Brownstein is a fox.

Yea I got your reference, I just really really like Carrie. But anyway, thanks to the OP i've been listening to SK all morning. too bad they're on "hiatus." there was a post on their website about a retrospective DVD about them, anyone heard any news on that?
